摘要
为了进一步探讨环胞霉素A (CSA)逆转白血病细胞耐药的机理 ,采用溴化四氮唑兰法(MTT)测定K5 62和K5 62 /ADM白血病细胞系对化疗的体外反应性 ,以脱氧核苷酸末端转移酶介导的缺口末端标记法 (TUNEL)及琼脂糖凝胶电泳检测细胞凋亡 ,台盼蓝拒染法测细胞存活率。结果发现CSA在 1.0mg/L水平对两种白血病细胞存活无明显影响 ,但能够增强其对化疗药物的敏感性 ,促进化疗药物诱导的白血病细胞凋亡 ,当浓度大于 1.0mg/L时单独CSA也能杀伤白血病细胞并诱导其凋亡。结论提示 ,CSA可以通过促进细胞凋亡逆转白血病细胞的耐药 。
To explore the mechanism of CSA in the reversal of drug-resistance of leukemia cells, K562 and K562/ADM cells were used for in vitro study. Drug sensitivity of cells was evaluated by MTT assay and cell survival by trypan blue exclusion. Apoptosis was detected by terminal-deoxynucleotidyl transferase mediated d-UTP nick end labeling (TUNEL) and agarose gel electrophoresis. The results showed that 1.0 mg/L CSA alone have no effect on the survival of K562 and K562/ADM cell lines, but it could enhance the chemotherapy sensitivity of the 2 cell lines, and accelerate the apoptosis induced by ADM. These effects were stronger in K562/ADM cell line, when the concentration of CSA>1.0 mg/L, CSA itself could kill leukemia cells and induce the apoptosis of the 2 cell lines. It was concluded that CSA can reverse leukemia cells′ drug-resistance by enhance apoptosis of the leukemic cells, CSA itself can induce leukemia cells′ apoptosis and kill leukemia cell too.
