摘要
目的 :寻找适合临床实际需要的逆转多药耐药的方法。方法 :用 MTT法和流式细胞仪测定技术研究环孢菌素 A(Cs A)、他莫西芬 (TAM)及α-干扰素 (IFN-α)单独应用和半量联合应用逆转裸鼠高致瘤性多药耐药人白血病细胞系 K5 6 2 - n/ VCR的作用。结果 :Cs A、TAM、IFN -α 3种药物单独和联合应用对 K 5 6 2 - n/ VCR细胞系的多药耐药均有逆转作用 ,Cs A单用逆转效果远远强于TAM及 IFN -α,Cs A>IFN -α>TAM;两两半量联合效果视不同组合而不同 ,三者半量联合应用比两两半量联合逆转效果强 ,而两两半量联合及三者半量联合应用的逆转效果与单独应用 Cs A相似 ,优于单独应用 TAM和 IFN-α。 Cs A使多药耐药细胞系 K 5 6 2 - n/ VCR细胞内 DNR含量增加 ,TAM及IFN-α不改变 K5 6 2 - n/ VCR细胞内 DNR含量。结论 :Cs A、TAM及 IFN -α 3种药物单独和联合应用对 K5 6 2 - n/ VCR细胞的多药耐药均有逆转作用 ,联合应用逆转作用比单独作用强 ,将 Cs A+TAM两药联合或 Cs A+TAM+IFN-α三药联合与化疗药物同时应用可能更有利于提高
Objective:To search new method to reverse MDR that can be applied in clinical practice.Methods:It is studied the reverse role of cyclosporin A(CsA), tamoxifen (TAM) and interferon α (IFN α) in a multidrug resistant leukemia cell line(K562 n/VCR) with high tumorigenicity in nude mice by MTT method and flow cytometer.Results:Multidrug resistance can partially reversed by addition of CsA, TAM, and IFN α. The intensity of this effect could be ordered as CsA>IFN α>TAM. DNR intracellular accumulation was partially reversed only by addition of CsA. The combination of either half dose CsA and TAM or half dose CsA, TAM and IFN α had more stronger effect than the full dose of each one. But the reversal effectiveness were weaker than that of all the modulators′ combination.Conclusion:The reversal agents (CsA, TAM and IFN α) have obvious effects on the MDR cell line. Combination recipes are more favored for higher effectiveness. It is suggested that combination of CsA +TAM or CsA+IFN α+TAM with anticancer drugs therapy should be applied in the MDR patients.
