姜黄素对K562/A02细胞上P-gp的表达及其功能的影响 被引量:5

摘要  目的: 观察姜黄素(curcumin, Cur)对人红白血病多药耐药(mμLtidrugresistance, MDR)细胞系K562 /A02细胞上P gp蛋白表达及功能的影响。方法: 用MTT比色法测定Cu作用后K562 /A02细胞对多种化疗药物敏感性的变化; 用流式细胞仪测定Cur对K562 /A02细胞内柔红霉素 (DNR)潴留的影响; 用RT PCR法检测Cur作用后K562 /A02细胞中mdr 1mRNA的表达。结果: Cur能增强多种化疗药物对K562 /A02细胞的毒性作用。明显提高K562 /A02细胞内DNR的浓度(P<0. 01)。不同浓度的Cur作用后, K562 /A02细胞中mdr -1mRNA的表达逐渐降低, 其中 2. 5mg/LCur处理组与未处理组相比较差异显著(P<0. 01)。结论: Cur能部分逆转K562 /A02细胞的耐药性, 且呈浓度依赖性。Cur的作用机制主要与下调mdr- 1mRNA的表达, 导致细胞膜P gp的表达减少有关。
