摘要
目的 探讨高压氧(hyperbaric oxygenation,HBO)对移植肾慢性排斥反应的防治作用及其机制。方法 以22只雌性F344大鼠为供体,22只雌性Lewis大鼠为受体,应用显微外科技术制作移植肾慢性排斥反应大鼠模型,将受体大鼠随机分为治疗和对照两组。治疗组 11只接受 HBO治疗,治疗压力为 0.2 MPa,每天 40 min;对照组 11只不接受 HBO治疗。移值后 24周处死动物,对移植肾组织行肾功能、组织学、免疫组化和反转录聚合酶链反应(RT-PCR)检测。结果 对照组蛋白尿明显多于治疗组,肌酥清除率明显低于治疗组,肾间质纤维化、肾小球硬化、动脉血管内膜增厚较治疗组严重。治疗组移植肾T细胞、单核/巨噬细胞浸润较对照组轻,但无统计学意义;而PDGF-AA表达水平较对照组明显低,其差异有统计学意义。结论HBO在预防或减轻同种移植肾慢性排斥反应中能起重要的作用。
Objective To explore the efficacy of hyperbaric oxygenation (HBO) on preventing renal allograft chronic rejection. Methods 22 female Lewis rats received kidney transplantation from fe-male F344 donors. Micro-surgery technique was used in transplant operation. All of the recipients were divided into two groups. 11 recipients were treated with HBOCO. 2 MPa 40 min each day) as treatment group. Other 11 recipients with no treatment as control group. 24 weeks after transplantation, all re-cipients were killed and renal functions were evaluated. Histologic, immunohistological and RT-PCR analysis were performed in kidney allografts. Results Compared to treatment group, the controls had significantly worse proteinuria; lower creatinine clearance rate; more severe tubulo-interstitial fibrosis and infiltration, glomerulosclerosis and arterial intima thickening. The infiltration of T-cells and mono-cytes/macrophoges in the allograft kidney of treatment group was less than that in the controls but without stat istical significance. More expression of PDGF-AA mRNA existed in the controls and was statistically significant as compared to treatment group. Conclusions HBO could play important role in preventing or reducing chronic allograft rejection.
