摘要
用塑料光纤将发光体系和发光仪的检测系统连接起来,通过光纤将反应的光传至化学发光仪,化学发光仪的检测系统可以灵敏地检出发光强度。在最佳工作条件下,NO浓度与相对化学发光强度之间有良好的线性关系,其线性范围为8.5×10-12mol/L至1.7×10-10mol/L,检出限为1.7×10-12mol/L。用该系统检测了加脂多糖、加N(G)-甲基-L-精氨酸、不加任何物质作对照三组培养细胞释放NO的水平。结果发现,NO水平由高到低依次为:LPS组>对照组>L-NMMA组。
A fiber-optic sensing technique of nitric chide(NO)based upon the chemiluminescence (Cl) reaction was developed ini- tially.The light emission is due to NO reacting with luminol-H2O2.Under these optimum conditions, the Cl value is proportional concentration NO in the range for 8.5× 10-12 MOL/L to 1. 7 × 10-10'mol/L, the detection limit is 1 .7 ×10-12mol/L.The NO levels of three groups of cultrued cells which were cultured respectively in medium 1640 with lipopolysaccharide (LPS), medium 1640 with L- NMMA,and medium 1640 as control. Experiment results showed; The order of NO concentrations is LPS group > controlled group>L- NMMA group.
