摘要
目的 用高效毛细管电泳法 ,对党参样品所含的活性成分中 4种有机酸类成分进行分离与测定。方法 采用高效毛细管区带电泳模式 ,二极管阵列检测器 ,在 2 0 9nm、2 5 4nm、30 8nm波长处进行定量测定。结果 最佳电泳条件为 :石英毛细管柱 (5 0 μmI.D .× 45cm) ,缓冲溶液为 2 0 %乙腈、2 5mmol/L硼砂溶液 (pH =9.12 ) ;分离电压为 18kV ,温度 2 8℃ ;样品和标准品在 2 0min内得到基线分离。结论 本方法简单、快捷、灵敏 ,当严格控制实验过程中的每个环节时 ,可以得到良好的准确性和重现性 。
Objective To study a new high performance capillary electrophoresis (HPCE) for quantitative determination of organic acids in the extract of Codonopsis pilosulae.Methods The contents of 4 organic acids were determined by using capillary zone electrophoresis (CZE) model. Diode arrary detector was set at 209 nm, 254 nm, and 308 nm wave lengths respectively.Results The best HPCE conditions were as follows: a fused silica capillary tube l eff =45 cm, L=53.5 cm, i.d.=50 μm; the background electrolyte system selected being 20% CAN mmol/L borate (pH 9.12); injection speed being 50 mbar/4s; applied voltage 18 kV; temperature being 28℃; and run time being 20 min. Samples and standards were entirely separated in less than 29 min.Conclusion This method is a simple, quick and effective tool for quality control studying herbal Codonopsis pilosulae medicine.
