摘要
目的 应用单细胞凝胶电泳 (SCGE)方法体外检测二硫化碳 (CS2 )染毒对人外周血淋巴细胞DNA损伤、损伤程度以及有无剂量 -效应关系。方法 将未处理的人外周血淋巴细胞作为阴性组 ,用H2 O2染毒的人外周血淋巴细胞作为阳性对照组 (5 0 μmol/LH2 O2 染毒 ) ,用不同浓度CS2 处理的人外周血淋巴细胞设为 4个剂量组 (5 0 0 μmol/L组、10 0 0 μmol/L组、2 5 0 0 μmol/L组、5 0 0 0 μmol/L组 ) ,进行SCGE实验。结果 随CS2 染毒浓度增加 ,淋巴细胞DNA损伤加重 ,表现在彗头直径变小 ,分别为 (5 .0 4± 0 .47)、(4 .95±0 .6 5 )、(4 .87± 0 .74)、(4 .92±0 .5 1) μm ,与阴性对照组比较 ,差异均有显著性 (P <0 .0 1) ;彗尾长度增加 ,分别为(5 .78± 7.0 4)、(12 .0 4± 8.95 )、(16 .12± 8.86 )、(17.2 8± 8.6 3) μm ,与阴性对照组比较 ,差异均有显著性 (P <0 .0 1)。DNA拖尾率在 5 0 0 μmol/L组 (4 8.73% )明显增加 ,阴性对照组为 11.5 6 % ;10 0 0 μmol/L组后未见随剂量增加损伤率增加的趋势 ,依次为 73.6 0 %、84.40 %、86 .86 % ,经 χ2 检验 ,6个组之间差异有显著性 (P <0 .0 1)。结论 CS2 体外染毒对人外周血淋巴细胞DNA有一定损伤 ,但CS2 染毒到达一定浓度后 。
Objective To detect the damage of DNA,the degree of DNA damage and the dose effect of human lymphocyte induced by CS 2 in vitro . Method Using the fresh human peripheral blood lymphocyte as the negative control group,the H 2O 2 50 μmol/L treated human peripheral blood lymphocyte as the positive control group,the different concentration CS 2 exposed human peripheral blood lymphocyte as four dose groups(500 μmol/L,1 000 μmol/L,2 500 μmol/L,5 000 μmol/L),DNA damage was detected by single cell gel electrophoresis(SCGE). Results When the concentration of CS 2 was increased,the damage of the DNA in the lymphocyte was aggravated.The head diameter of the comet was diminished,the length of the comet was increased.The rate of DNA tail increased significantly in the groups of 500 μmol/L and 1?000 μmol/L,however,not so obviously increased in the groups of 2?500 μmol/L and 5?000 μmol/L. Conclusion CS 2 exposure in vitro may have certain injury to human peripheral blood lymphocyte DNA,but the damage was not definitely aggravated when CS 2 dose reached a certain extent.
