摘要
目的 观察N 乙酰 L 半胱氨酸 (NAC)对体外循环 (CPB)血清诱导的培养血管内皮细胞细胞间粘附分子 1(ICAM 1)的表达及对多形核粒细胞 (PMN)粘附的影响 ,探讨NAC对体外循环后全身炎症反应的作用及机制。方法 以CPB血清加NAC处理培养人脐静脉内皮细胞 (HUVEC) ,采用中性粒细胞 血管内皮细胞粘附模型研究HUVEC对PMN的粘附作用 ,以免疫组化方法观察培养血管内皮细胞表面ICAM 1的表达。结果 NAC加入CPB血清共同处理HUVEC ,能抑制CPB血清诱导的HUVEC对PMN的粘附增加 ,下调CPB血清诱导的HUVEC表面ICAM 1表达的增高。结论 NAC通过抑制内皮细胞表面ICAM 1的表达 ,可抑制体外循环诱导的PMN与血管内皮细胞的粘附 ,从而可能在体外循环后全身炎症反应的防治中发挥作用。
Objective To investigate the effect of N acetyl L cysteine (NAC) on the intercellular adhesion molecule 1 (ICAM 1) expression and the adhesion of polymorphonuclear (PMN) cells to the cultured endothelial cells induced by cardiopulmonary bypass (CPB) serum. Methods The PMN endothelium adhesion model was employed to study the adhesion of PMN to human umbilical vein endothelial cells (HUVEC) cultivated with CPB serum containing NAC. The expression of ICAM 1 in endothelial cells was detected with immunohistochemical methods. Results NAC inhibited CPB serum induced HUVEC adhesion to PMN and down regulated the ICAM 1 expression which had been increased by CPB serum on HUVEC surface. Conclusion These data indicate that NAC could inhibit PMN endothelium adhesion by down regulating ICAM 1 expression on endothelial cells induced by CPB serum.
