妊高征血清对脐静脉内皮细胞影响的研究 被引量:4

摘要 目的 利用妊高征患者血清致伤培养的正常脐静脉内皮细胞 ,观察致伤后内皮细胞分泌功能的变化。方法 培养后的脐静脉内皮细胞用中、重度妊高征患者的血清为干预条件 ,共同培养后 ,测定NO2- NO3- 、LDH、SOD、MDA的含量变化。结果 ①LDH的活性、MDA的含量 :中、重度妊高征组 2 4h点及重度妊高征组 12h点均明显高于 6h时间点(P <0 .0 5 ) ,12、2 4h两时间点均明显高于对照组相同时间点 (P <0 .0 5 ) ;②SOD含量 :重度妊高征组 12、2 4h点均明显低于6h点 (P <0 .0 5 )。中、重度妊高征组 2 4h点及重度妊高征组 12h点均明显低于对照组相同时间点 (P <0 .0 5 ) ;③NO2 - NO3- 含量 :中、重度妊高征组内 2 4h点均明显低于 6h点 (P <0 .0 5 )。中、重度妊高征组的 2 4h点及重度妊高征组 12h点均明显低于对照组相同时间点 (P <0 .0 5 )。 Objective To observe the changes in secretory functions of cultured umbilical vein endothelial cells injured by serum from pregnancy induced hypertension (PIH). Methods Endothelial cells isolated from the umbilical veins from moderate or severe PIH women (PIH group, n =10) and normal pregnant women (control group, n =8) were incubated at 37 ℃ in humidified air with 5% CO 2. Cell culture fluids were obtained at 3 and 4 d of primary culture. Contents of nitric oxide (NO), malondialdehyde (MDA), superoxide dismutase (SOD), lactate dehydrogenase (LDH ) in the cell culture fluids were measured. Results LDH activity and MDA contents in the cell culture fluids in PIH group were significantly higher than those in the control group ( P <0.05). SOD activity and NO contents in the cell culture fluids in PIH group were significantly lower than those in the control group ( P <0.05). Conclusion Injury occurs in the umbilical vein endothelial cells from PIH.
