摘要
目的 观察 5/6肾切除大鼠肾脏细胞凋亡及凋亡相关基因 Bax、Bcl-2、Fas和 Fas-L的表达变化及其意义。方法 SD大鼠行5/6肾切除,术后6周、12周采集标本,除常规光镜、电镜观察外.利用TUNEL及流或细胞术,观察肾脏细胞凋亡及凋亡相关基因表达的变化,并结合肾功能变化分析肾脏细胞凋亡及凋亡相关基因表达与肾小球、肾小管及间质病变的关系。结果5/6肾切除大鼠肾脏细胞凋亡增加,凋亡相关基因表达增强,这些变化与肾脏形态和功能变化相平行 。结论 肾脏细胞凋亡及凋亡相关基因表达的变化可能参与肾小球硬化、肾小管萎缩及间质纤维化的病理过程。
Objective To investigate the role of apoptosis and expression of apoptosis-related genes in renal sclerosis. Methods Remnant kidneys were produced by 5/6 nephrectomy. The tissues of remnant kidneys were used to detect apoptosis and the cxpression of Bax, Del-2, Fas and Fas-L by TUNEL and flow cytometry. Results The number of apoptotic cells was more and the expression of Bax, Dcl-2, Fas and Fas-L was higher in the remnant kidneys. The successive changes of thase parameters were parallel to the morphological and functional alterations of the kidneys. Conclusion Apoptosis and the related genes may be involved in the development of chronic renal sclerosis.
