三七总皂甙对大鼠脊髓损伤后HSP_(70)阳性神经元表达的影响 被引量:7

摘要 目的 :探讨脊髓继发性损伤的病理生理机制及三七总皂甙 ( Panaxnotoginseng saponins,PNS)防治该病的作用机理。方法 :以 Wistar大鼠 65只为研究对象 ,分为空白组、假手术组、溶媒对照组、三七总皂甙( PNS)组 ,各实验组按 2、6、1 2、2 4小时相点观察 ,Allen's脊髓损伤模型 1 0 0 g· cm致伤脊髓。各组在相应时相点取脊髓组织 ,采取免疫细胞化学方法观察脊髓组织热休克蛋白 70 ( Heat Shock Protein 70 ,HSP70 )阳性神经元的表达变化。结果 :脊髓 HSP70 阳性神经元表达 :PNS组 >溶媒对照组 >假手术组 >空白组。结论 :HSP70 参与了脊髓继发损伤的病理生理机制 ,PNS可能通过诱导脊髓组织 HSP70 神经元的表达而起保护作用。 Objective:In order to investigate the effect of the heat shock protein 70(HSP 70) following secondary spinal cord injury, and to determine whether the Chinese medicine panax notoginseng saponins(PNS) has the protection through affecting the chang of HSP 70.Methods:Wistar rats were used to observe the expression of HSP 70 in the nervous cell of spinal cord with immunocytochemical methods. The Wistar rats were divided into blank group, control group, solute group and PNS group(control group, solute group and PNS group were divided into two, six, twelve, twenty-four hours group again). Allen's technique 100g·cm onto surgically exposed dorsal surface of spinal cord. Results:The expression density of the HSP 70 positive neuron of spinal cord: the PNS group > the solute group > the control group > the blank group. Conclusion:HSP 70likely partake into the mechanism of pathology and physiology in the process of the secondary injury of spinal cord of rats. PNS can induce the expression of HSP 70 in the prevention and cure of the secodary injury.
