摘要
目的 观察大鼠脑缺血后缺血区内皮素 1基因表达的变化和部位 ,以及丹参对内皮素 1基因表达的影响。方法 采用线栓法制成大鼠大脑中动脉缺血模型。应用点杂交的方法 ,动态观察大鼠持续性脑缺血前和缺血后0 .5、1.5、3、6、12、2 4、48和 72h皮层和尾壳核内皮素 1mRNA水平的变化 ;应用原位杂交的方法 ,观察了脑缺血后 2 4h大脑表达ET 1mRNA的部位及细胞类型 ,以及丹参的影响。结果 缺血侧皮层和尾壳核ET 1mRNA水平于缺血6h开始明显升高 ,于缺血 2 4~ 48h达高峰。在缺血侧大脑中动脉分布区有广泛ET 1mRNA表达。在ET 1基因表达阳性的细胞中 ,既有血管内皮细胞 ,又有神经细胞。丹参预防组ET 1基因表达增强 ,但低于对照组。结论 脑缺血可诱发ET 1基因表达增加 ,导致ET 1合成增加。丹参能部分抑制脑缺血后ET 1基因的表达 ,这可能是其治疗缺血性脑血管病的机理之一。
Objective To investigate the endothelin 1 gene expression and effect of Radix Salviae Miltiorrhizae(RSM) in the ischemic area after cerebral ischemia.Methods The focal cerebral ischemia models of rats (n=33) were made with thread embolism of left middle cerebral artery(LMCA).The endothelin 1(ET 1) mRNA levels in the cerebral cortex and caudoputamen before and at 0.5,1.5,3,6,12,24,48 and 72 h of ischemia were investigated with dot blot hybridization.24 h after LMCA occlusion,the ET 1 mRNA expression was investigated by in situ hybridization.Results It was shown that the levels of ET 1 mRNA in the cortex and caudoputamen of the ischemic side were enhanced obviously at 6 h of ischemia and reached the highest levels at 24~48 h.The ET 1 mRNA was extensively induced in the area supplied by the MCA.The ET 1 gene expression was found in both vessel endothelial cells and neuronal cells.In RSM protected rats,the ET 1 gene expression was also significantly increased,but was lower than that of controls.Conclusions The results indicated that the ET 1 gene expression in the brain can be induced by ischemia,leading to increase in synthesis of ET 1.The protective effect of RSM on cerebral ischemia may relate to its inhibitory effect on ET 1 gene expression.
