摘要
本文报告我国登革2型04株包膜蛋白基因的核苷酸及氨基酸序列,并与登革1、3、4型及其它2型毒株如几内亚C株(NGC)、牙买加株1409(JAM)、候选疫苗株S1以及马来西亚当地流行株M1(登革出血热)、M2(登革休克综合征)。 M3(登革热)进行比较。结果表明D_2-04株E蛋白基因的核苷酸序列与NGC,JAM和S1的同源性分别为95.0％、97.4％和90.0％;与M1、M2、M3的同源性分别为93.3％、92.1％和94.3％;与其它登革血清型的同源性大约为65％。登革2型04株蛋白的氨基酸序列与NGC、JAM、S1、M1、M2、M3的类似性分别为96.8％、97.2％、91.4％、95％、95.6％和94.3％。与其它登革血清型氨基酸类似性大约为62％～68％;与其它黄病毒的类似性范围为43％～47％。推断的氨基酸序列显示出7个保守的半胱氨酸残基及两个潜在的糖基化位点,分别位于Asn-67和Asn-153位。
We reported the nucleotide and encoded amino acid sequences of the envelope (E) protein gene of a dengue-2(DEN-2) virus strain 04, isolated from a patient in Hainan, China in 1985. The sequences were compared with the corresponding sequences from DEN-1, DEN-3, DEN-4 and other DEN-2 viruses: the prototype New Guinea G virus (NGG), the Jamaican (JAM) strain 1409, the candidate vaccine strain PR159 (S1) from Puerto Rico and three Malaysia isolates (M1, M2 and M3). The nucleotide sequence encoding the E protein was determined by dideoxyribonucleotide chain termination method. Comparison of D_2-04 E nucleotide sequence with the corresponding sequence from NGC, JAM and S1 revealed similarities of 95.0％, 97.4％ and 90.0％ respectively. The other DEN-2 viruses showed similarities from 92.3％ (M1) to 94.3％ (M3), while the other dengue serotypes showed similarities of about 65％. Comparison of the deduced amino acid sequence of D_2-04 E protein with the corresponding sequence from NGC, JAM, S1, M1, M2 and M3 revealed similarities of 96.8％, 97.2％, 96.4％, 95.0％, 95.6％ and 94.3％ respectively. The similarities with other dengue serotypes were 62—68％, while those with other flaviviruses ranged from 43.8％ (yellow fever virus) to 47％ (JE). The deduced amino acid sequence revealed seven cysteine residues conserved within the DEN 2 serotype, as well as two potential glycosylation sites at Asn-67 and Asn-153.
