摘要
目的 :探讨黄芪注射液和川芎嗪注射液配伍防治心肌缺血再灌注损伤 (MIRI)的作用机制。方法 :在体兔结扎冠状动脉心肌缺血再灌注损伤模型 ,随机分为对照组、黄芪注射液组 (益气组 )、川芎嗪注射液组 (活血组 )、黄芪和川芎嗪注射液配伍组 (益气活血组 )。术中动态监测心电图并留取静脉血检测不同时期血清中乳酸脱氢酶 (L DH)、肌酸激酶 (CK)、丙二醛 (MDA)含量和超氧化物歧化酶 (SOD)、一氧化氮 (NO)、一氧化氮合酶 (NOS)活性。结果 :治疗各组与对照组比较 ,心电图损伤较轻 ,能减轻 L DH、CK、MDA和 SOD的变化 ,以益气活血组最为明显 ,与对照组比较多数有显著性差异 (P<0 .0 5或 P<0 .0 1)。益气组和益气活血组 NO和 NOS活性升高 ,而活血组 NOS活性降低。结论 :黄芪和川芎嗪注射液均有保护 MIRI的作用 ,两药作用机制略有不同 ,但两药伍用具有协同互补作用。
Objective:To investigate the mechanism of Astragalus membranaceus injection(黄芪注射液) combined with Ligustrazine injection(川芎嗪注射液) in protecting and treating myocardial ischemiareperfusion injury (MIRI).Methods:Thirtyfour rabbit models with MIRI established by clamping left branch of their coronary artery were divided randomly into control group and three treated group (Astragalus membranaceus,Ligustrazine,Astragalus membranaceus combined with Ligustrazine).ECG was dynamically observed.And the serum levels of creatine phosphokinase (CPK),lactate dehydrogenase (LDH),malondialdehyde (MDA) and the activities of superoxide dismutase (SOD),nitric oxide (NO),and nitric oxide synthetase (NOS) were determined at the different stage.Results:In all the treated groups in comparison to control group the damage to ECG was lighter and the changes in LDH,CPK,MDA and SOD were lighter too,meanwhile most of them had significant differences (P<0.05 or P<0.01).Among them in Astragalus membranaceus combined with Ligustrazine the difference was the most significant.Otherwise in Astragalus membranaceus group and Astragalus membranaceus combined with Ligustrazine group the activities of NO and NOS were increased but in Ligustrazine injection group the activities of NOS decreased.Conclusions:Both Astragalus membranaceus injection and Ligustrazine injection possess protective effects on MIRI.But the mechanisms of their actions are a little bit different,furthermore there are synergic and complementary actions if the two drugs are used together.
