摘要
目的 建立合适的以聚合的牛血红蛋白 (Hb)为基质的血液替代品的制备方法。方法 用乙醇醛聚合牛Hb ,用电泳和HPLC检测聚合程度、超滤去除大于相对分子质量 (Mr) 10 0 0 0 0 0的大分子和小于 10 0 0 0 0的小分子 ,用生理溶液平衡后 ,检测其生物学活性和流体力学特性。结果 牛Hb被有效聚合、超滤后 ,Mr超过 5 0 0 0 0 0的组分还占 4.6 % ,32 0 0 0的占 7.3% ,6 40 0 0~5 0 0 0 0 0的组分占 88% ;牛Hb在聚合后氧亲和力、血氧饱和度和溶液的溶氧量基本保持不变 ,其溶液黏度低 ,近似牛顿液体。结论 牛Hb经乙醇醛聚合后 ,Mr分布、生物学活性和流体力学特性理想 。
OBJECTIVE: To establish the suitable methods for the polymerization of bovine hemoglobin(Hb) and the manufacture of blood substitute. METHODS: Hb was polymerized with glycolaldehyde. The degree of polymerization was detected with SDS - PAGE and HPLC, The polymerized products between 100000 and 1000000 was separated and balanced with physiological solution by ultra - filtration. The poly Hb solution' s biological activity and hydrodynamics characteristics were measured. RESULTS: Bovine Hb was polymerized effectively. After ultra - filtration, the amount of polyHb between 64 000 and 500 000 reached to 88% of total polyHb. Its oxygen affinity, oxygen saturation degree and the amount of soluble oxygen in the polyHb solution were similar to whole blood. The polyHb solution was Newton' s liquid. CONCLUSION: In terms of the scale of molecular size, the biological activity and the hydrodynamic characteristics, glycolaldehyde - polymerized -bovine Hb solution was suitable to act as a new potential blood substitute.
