摘要
运用正交试验理论 ,对L 乳酸高产突变株NAF 0 32的优化摇瓶发酵条件作了初步研究 ,确定了优化的发酵培养基和发酵条件 .优化发酵培养基为 (g/L) :葡萄糖 140 ,氯化铵 1,KH2 PO40 .3,MgSO4 ·7H2 O 0 .2 5 ,ZnSO4 ·7H2 O 0 .0 8;优化培养条件为 :34℃ ,摇床转速 2 0 0r/min ,装液量5 0mL(2 5 0mL容积三角瓶 ) ,一次性添加CaCO380 g/L调节pH值 .米根霉NAF 0 32的摇瓶发酵L 乳酸积累量可达 94 .2 8g/L .
The optimal production medium and fermentation conditions of a mutant strain of NAF 032 with a high and stable yield of L lactic acid were studied by means of an orthogonal design. From the shake flask experiments, the optimal fermentation medium consisted of the following composition(g/L):glucose 140, NH 4Cl 1,KH 2PO 4 0.3,MgSO 4·7H 2O 0.25,ZnSO 4·7H 2O 0.08. The optimal fermentation conditions were: 34 ℃,200 r/min, 50 mL in 250 mL total volume, CaCO 3 80 g/L. CaCO 3 was added to the broth to control the pH value. The yield of NAF 032 in the shake flask experiments can be up to 94.28 g/L in the optimal fermentation.
