摘要
为探讨姜黄素诱导急性髓系白血病 (AML)原代细胞凋亡的作用及其机制 ,采用流式细胞术作细胞周期分析并检测 Sub- G1 峰值 ;SABC法检测 Bcl- 2家族 Mcl- 1、 Bax和 Bak蛋白表达。结果显示姜黄素不影响初治 AML 原代细胞细胞周期进展 ,但使 Sub- G1 峰值升高 (P<0 .0 5 )。在初治 AML 原代细胞 ,同样处理可下调 Mcl- 1表达、上调 Bax和Bak表达 ,且结果有显著性差异。提示姜黄素可诱导初治 AML 患者原代细胞凋亡 ,Bcl-
To investigate the apoptosis inducing effect and underlying mechanism of curcumin on primary first diagnosed acute myeloid leukemic (AML) cells, flow cytometric technique was used to do cell DNA analysis for measuring cell cycle and Sub G 1 peak value. The protein expression of Bcl 2 family anti apoptotic member Mcl 1 and pro apoptotic members Bax, Bak were detected by immunohistochemical staining assay (SABC). Results showed that in primary first diagnosed AML cells, there was no alteration of cell cycle progression with curcumin treatment. But the Sub G 1 peak was increased significantly after curcumin treatment at 25 μmol/L for 24 h ( P <0.05). The same treatment could down regulate Mcl 1 and up regulate Bax and Bak protein expression in first diagnosed primary AML cells. These results achieved significant difference. In conclusion, it was proved firstly that curcumin could induce apoptosis in first diagnosed AML cells. Regulation of expression of Bcl 2 family protein may be involved in apoptosis induction by curcumin for acute leukemic cells.
