摘要
目的 :观察姜黄素和阿霉素联合应用对体外培养的人肝癌细胞 SMMC- 772 1生长的影响。方法 :采用 MTT法应用 Webb系数判定 2药合用的效果 ,应用流式细胞仪和透射电镜观察药物对细胞周期分布及超微结构的影响。结果 :2种药物单独应用时随着药物剂量增加 ,其效应也增加 ;2药大剂量合用时可产生拮抗作用 ,小剂量合用时可产生协同作用。结论 :在体外 ,2 .5 mg/L~ 5 m g/L的姜黄素和 0 .12 5 mg/L~ 0 .5 mg/L的阿霉素联合应用 ,对 SMMC- 772 1具有协同抑制作用。
Objective:To Study the effect of curcumin and adriamycin on human hepatocarcinoma cell lines SMMC 7721 in vitro.Methods:Judging the effect of two drugs on cell growth,cell cycle distribution and characteristic ultrastructural alterations through MTT assay,webb coefficent,flow cell cycle and electron microscope were used.Results:In vitro,both curcumin and adriamycin could inhibit the proliferation of SMMC 7721 cells.Antagonism was produced when two drugs were used at a high concentration,and synergism was achieved when at a low concentration.Conclusion:In vitro,synergism was shown by using curcumin(2.5 mg/L~5 mg/L) and adriamycin(0.125 mg/L~0.5 mg/L).Curcumin can increase efficacy and decrease side effects of adriamycin.
