人肝肿瘤细胞株 Hep G2可应用于体外遗传毒理学试验等多种研究。但其培养难度较大 ,通过对 Hep G2细胞在不同培养条件下生长状态的对比 ,探讨了 Hep G2细胞培养增殖的最佳条件。结果表明在下列情况下细胞能顺利传代生长 :复苏时需 2 0 %胎牛血清 ,接种密度为 1× 10 4/ cm2 ,需在含 5 % CO2 环境下 ;培养时需 10 %胎牛血清 ,可在无 5 %
The human hepatoma line (HepG2) can be used in a variety of studies, but it is difficult to culture. The optimal culture conditions of HepG2 cells were studied by the contrast of their growth state under different culture conditions. The results demonstrated the cells could be subcultured smoothly under the following situation: (1) for resuscitation, 20 % fetal bovine serum was needed and the seeding density was 1×10 4 cells/cm 2 under humidified atmosphere of 50 % CO 2 ; (2) for culturing, 10 % fetal bovine serum was needed and humidified atmosphere in 5 % CO 2 was optimal.
