摘要
目的 :从分子水平研究BMP 2在黄韧带骨化过程中的作用机理。方法 :以rhBMP 2诱导的黄韧带骨化 (实验组 ,O组 )为研究对象 ,分别于诱导物植入后 2、4、6周取实验节段黄韧带 ,提取总RNA ,采用Dotblot和Northernblot方法检测其BMP 2mRNA的表达程度 ,设立PBS溶液 /明胶海绵植入为实验对照组 (EC组 ) ,未手术动物为阴性对照组(N组 ) ,pSBMP 2质粒为阳性对照组 (P组 )。结果 :Dotblot和Northernbloe放射自显影图像显示 :rhBMP 2植入 2周时 ,黄韧带内BMP 2mRNA表达程度最高 ,然后逐渐下降 ,到 6周时明显减弱 ,与病理组织学研究中所见的韧带组织中成纤维细胞软骨化生的活跃程度呈正相关。实验对照组和阴性对照组均未见BMP 2mRNA表达。图像灰度扫描示 :0 2组、0 4组和 0 6组BMP 2mRNA的表达程度分别为 0 .6 15± 0 .14 84、 0 .443± 0 .0 982、 0 .2 32± 0 .0 749。结论 :rhBMP 2诱导的骨化过程中黄韧带组织内存在不同程度BMP 2mRNA的表达 ,其表达水平与成纤维细胞软骨化生的活跃程度呈正相关 ;内源性BMP -2可能参与黄韧带骨化的启动和持续发展。
Objective:To study the mechanism of effection of BMP 2 in the course of ligamentum flavum ossification through gene analysis. Methods:Resect the tissue of ligamentum flavum of experimental segment in 2,4 and 6 weeks after implantation of rhBMP 2/gelform sponge composite (O group).Distract the total RNA from the pathological tissue. Detect the expression degree of BMP 2 mRNA using methods of Dot blot and Northern blot.Set up the impanting of PBS solution/gelform sponge composite as experimental control group(EC group). Animals without operation as negative control group (N group).pSBMP 2 plasmid as positive control group(P group).Results:Autoradiography of Dot blot and Northern blot showing that the expression of BMP 2 mRNA is the highest in the tissues resected from the animals 2 weeks after implanting of rhBMP 2. Then decrease gradually. The decreasing is obviously in 6 weeks. The changes of expression degree relate positively to the activity of chondrolization of fibroblast as shown in the study of pathology. There is no expression of BMP 2 mRNA in EC group and N group. The imaging dense scanning show that the values of expression degree of BMP 2 mRNA are 0.615±0.0184,0.443±0.0982 and 0.232±0.0749 seperatly in 2,4, and 6 weeks after implanting of rhBMP-2 Conclusions:There are different degrees of expression of BMP 2 mRNA in the tissues of ligamentum flavum during the ossification progression induced by rhBMP 2. The degree of expression relate positively to the activity of chondrolization of fibroblast. The internal BMP 2 may participate in the starting and continuing of ossification of ligamentum flavum.
