摘要
目的 制备日本血吸虫单克隆抗独特型抗体 (anti- id,Ab2 ) NP30的抗抗独特型抗体 (an-ti- anti- id,Ab3)。方法 IRS- NP30主动免疫 BAL B/ c小鼠 ,取其脾细胞与 SP2 / 0细胞融合 ,制备 an-ti- anti- id杂交瘤细胞株。结果 获得 1株稳定分泌单克隆抗体的细胞株 ,定名为 NP41。 NP41的 Ig同型为 Ig M。免疫组化显示 NP41与成虫的体膜、消化管上皮、子宫内膜上皮及虫卵内的毛蚴头腺、毛蚴膜呈阳性反应。结论 NP41为 NP30的 anti- anti- id,其识别的抗原即
Objective To fabricate anti anti idiotypic antibody (anti anti id,Ab 3) of monoclonal anti idiotypic antibody NP30 (Ab 2) of Schistosoma japonicum.Methods The hybridoma cell line of anti anti id was obtained by fusion of myeloma cells SP2/0 and spleen cells from BALB/c mice immunized actively by the IRS NP30. Results A hybridoma cell line secreting a monoclonal antibody, named NP41, was established. The immunoglobulin isotype of NP41 was determined as IgM. The NP41 reacted positively to outer layer membrane of body, the lining membrane tissue of gut cavity and epithelium of endometrium of adult worm, and head gland and membrane of miracidia in eggs ofSchistosoma japonicum by method of immunohistochemistry. Conclusion [WT5”BZ]NP41 is anti anti id of NP30 and the antigen distinguished by NP41 is the mimic antigen of NP30.
