摘要
本实验用抗疟原虫子孢子混合单抗MS1—5免疫同系小鼠获得一株抗独特型单克隆抗体(D3)。经竞争抑制ELISA、IFA以及放免中和抑制试验等证实:D3能模拟抗原CSP诱导特异性抗子孢子抗体的产生,使兔抗D3具有抗子孢子的抗体活性。反之,兔抗子孢子抗体又具有抗D3的活性,两者的相互结合既能被异种抗子孢子抗体所中和,又能被抗原所竞争。不仅如此,D3杂交瘤细胞表面的Ig还表现出于孢子抗原决定簇的抗体结合活性。表明D3抗体是一株具有抗原内影像的抗独特型抗体,即Ab2β。
Hybridoma (D3) secteing monoclonal anti-idiotype antibody against MS1-5 was produced by fusion of sp2/0 myeloma cells and spleen cells of syngeneic mice immunized with the mixture of MSl-5 D3 was identified with a series of experiments inclu-. ding competitive and neutralizing inhibition ELISA. D3 antibody could induce anti-sporozoite antibody when administered into rabbit and BALB/c mice, on the other hand, rabbit anti-sporozoite could react specifically with D3. It was found that D3 could competitively inhihibit the binding of sporozoite with antisporozoite of various origin, and also neutralized by anti-sporozoite, D3 hybridoma cells could be stained specifically with anti-sporozoite antibody These results showed that anti-idiotype antibody produced by D3 hybridoma is the anti-idiotype dearing internal image of CSP epitope, i. e Ab2 β.
