摘要
目的诱变、筛选γ 亚麻酸 (GLA)高产菌株。方法采用 5 氟尿嘧啶、紫外线、氯化锂复合诱变及自然分离的方法 ,对拉曼被孢霉AS 3.34 13进行诱变筛选。结果获得一株GLA高产突变株F5。该菌株GLA产量较原始菌株提高了 30 0 %。传代实验证明该菌株遗传性质稳定。经对其发酵培养基及发酵条件的优化 ,该菌株GLA生产能力可达 115 3.6 3mg/L ,较原始菌株提高 335 .87%。结论拉曼被孢霉GLA高产突变株F5已达工业生产菌株要求。
PurposeThe aim is to breed a highly productive γ-linolenic acid mutant strain.MethodsThe original strain Mortierella ramanniance AS 3.3413 was treated by 5-FU,UV,LiCl as mutanting agent.ResultsA highly productive γ-linolenic acid mutant strain F 5 was successfully obtained.The γ-linolenic acid yield of mutant strain F 5 was increased by 300% as compared with that of the original strain.Pass-generation test showed that the heredity character of mutant strain F 5 was stable.After the optimal medium and culture condition was established, the yield of γ-linolenic acid was up to 1 153.63mg/L,and was increased by 335.87% as compared with that of the original strain.ConclusionThe mutant strain can be used for the industrial production of GLA.
