用高大毛霉生产γ-亚麻酸的原生质体制备与诱变 被引量:2

摘要 利用双酶法对产γ-亚麻酸的高大毛霉进行原生质体制备与诱变,结果表明:当w(纤维素酶)=2.0%,w(蜗牛酶)=0.5%,处理培养10h的菌丝,酶解时间3h,原生质体形成率>89.23%;将所获得的原生质体直接进行紫外线诱变,得到的突变株经发酵培养,获得一高产菌株,其油脂产率为7%,γ-亚麻酸质量分数可达41.5%,γ-亚麻酸产量比原始菌株提高了31.3%. The protoplast formation of original strain Mucor mucedo which produces γ-linolenic acid by using a combined enzyme system was studied.The results showed that,it was beneficial under the conditions of using 10 h culitivated cell,w(cellulose enzyme)=2.0%,w(snail enzyme)=0.5% for 3 h.The ratio of protoplast formation was more than 89.23%.Mutant strains were treated by protoplast-UV inducing method,and a highly productive γ-linolenic acid mutant strain was obtained,of which the oil content and γ-linolenic content were 7% and 41.5%.Its γ-linolenic acid yield of it was increased by 31.3%,as compared with that of the original strain.
