鼠疫耶尔森菌中国分离株菌体脂肪酸成分分析 被引量:9

摘要 目的 探讨利用脂肪酸组成对中国鼠疫耶尔森菌菌株进行分型的可能性 ,获得中国鼠疫耶尔森菌菌株脂肪酸组成的本底资料。方法 选择历年来从我国不同疫源地分离到的 5 8株鼠疫耶尔森菌进行了菌体脂肪酸成分分析 ,并利用STATISTICA统计软件进行实验菌株的聚类分析。结果 中国鼠疫耶尔森菌菌株的主要脂肪酸成分为 16∶0酸 ,环式 17∶0酸 ,3 羟基 14∶0酸和ω7c 16∶1酸以及十八碳单烯酸 ,与MIDI公司Sherlock系统的菌库中的数据存在一些差异。结论 实验菌株的脂肪酸成分极为接近 ,聚类图不能充分体现分离株在分离时间、地点以及生物型别上的差异 ,表明脂肪酸分析目前不适于鼠疫耶尔森菌的分型。根据实验结果 。 Objective To exploite the possibility of utilizing CFAs (cellular fatty acids) information in typing Yersinia pestis ; to collect CFAs background data of Yersinia pestis strains in China. Methods 58 Yersinia pestis strains, including 51 wide strains isolated in different regions in China, 6 lab mutant strains and one vaccine strain EV76, were subjected to CFA study. All experimental strains were inoculated on TSBA (trypticase soy broth agar) plates for 24 hours. After that, cells were harvested, saponificated, methylated and extracted to provide FAMEs(fatty acids methyl esters) for gas chromatography analysis. Based on the CFAs data matrix, dendrogram of 58 experimental strains was generated by Statistica 5.0 software package. Results Major CFAs of experimental strains are 16∶0, cyclo 17∶0, 3 OH 14∶0,ω7c 16∶1 and monounsaturated 18 carbon acids, which account for more than 70% of the total CFAs. Conclusion Dendrogram of experimental strains reveals a highly homogeneity among these Yersinia pestis isolates,which suggests that CFAs analysis can not be applied as a competent tool in typing Yersinia pestis strains. All the experimental strains are identified as Yersinia pseudotuberculosis by MIDI sherlock system, which were made up by CFAs difference between experimental strains and Sherlock standard library strains.According to this finding, a CFAs database for Chinese isolates of Yersinia pestis has been developed.
