摘要
我们建立了用UDG(尿嘧啶糖基化酶)防PCR产物污染,以两个不同基因为靶序列扩增检测同一病原菌和用微孔板杂交-酶联显色检测PCR产物的UDPE(UDG-Duplex PCR-EIA)技术,并用于鼠疫耶尔森氏菌的检测与鉴定。结果表明,该系统可以特异地检测和鉴定鼠疫耶尔森菌,检测灵敏度可以达到1CFU/ml。
A Method for detecting and identifying Yersinia pestis, called UDPE (UDG - Duplex PCR -EIA) , was established. The advantages of this method were demonstrated by employing uracil de-oxynucleotide glycosidase(UDG) for anti - carryover of PCR products, using two sets of primers targeting two different genes for detecting and identifying one bacteria to avoid the false - negative results caused by genetic variations and detecting PCR products by microplate hybridization and enzyme im-munoassay (EIA) . The fasibility of this method in detection and identification of Yersinia pestis showed it a specific and sensitive technique. 1 CFU/ml could be detected.
