摘要
目的 比较DL -PLA、 2 0 % β -TCP/DL -PLA制成的三维多孔材料降解特点 ,选择有利于骨缺损修复的材料。 方法 应用浇铸盐析技术分别将DL -PLA、 2 0 % β-TCP/DL -PLA制成三维多孔结构 ,通过体外降解实验观察测量并对比两种材料的分子量、重量、生物力学、pH值的动态变化。结果 在降解过程中DL -PLA材料 pH值在 3周内轻度下降 ,而后明显下降 ,12周时仅为 2 94。 2 0 % β -TCP/DL -PLA材料在 12周的降解过程中 pH值十分稳定 ,12周时为 6 6。 2 0 % β -TCP/DL-PLA在 0~ 2周分子量变化较大 ,从 3940 0下降为 2 82 0 0 ,2周后下降较平缓 ,12周时为 12 0 0 0 ,而DL -PLA在 0~ 2周分子量变化极大 ,从 3940 0下降为 10 90 0 ,12周时为 2 940。 2 0 % β-TCP/DL -PLA重量变化率 0~ 4周时变化较小 ,4周后下降加快 ,12周时为初始重量的 6 0 %。DL -PLA与之相似 12周时为初始重量的 5 5 %。初始强度 2 0 % β -TCP/DL -PLA为 1 72MPa,DL -PLA为 1 35MPa ,两者有显著差别 (P <0 0 5 )DL -PLA在降解过程中DL -PLA组 0~ 4周下降极小 ,4~ 6周下降较快 ,12周为 0 13MPa ,2 0 % β -TCP/DL -PLA在 0~ 6周下降极小 ,6~ 8周下降较快 ,12周时为 0 2 2MPa。 结论 三维多孔 2 0 % β -TCP/DL -PLA作为一种?
Objective To select a better bone repair material to heal bone defect between DL-PLA and 20% β-TCP/DL-PLA which were prepared as 3-D porous.Methods A particulate leaching method was developed to prepare 3 D porous DL-PLA and 20% β-TCP/DL-PLA.In vitrio,two types of material were put into water(pH7 0).The pH,molecular weight,weight and biomechanics were observed during the degradation in 12 weeks.Results During degradation the pH and molecular weight of DL-PLA changed obviously,at the end of 12 weeks the pH is 2 94,molecular weight is 2940 and the pH of 20%β TCP/DL-PLA change little at the end of 12 weeks is 6.6,molecular weight is 12000,which shows a significant difference(P<0 05).At the end of 12 weeks the weight of DL-PLA remains 60% and 20% β-TCP/DL-PLA is 55%,there is no significant difference between them(P<0 05).The prime biomechanic strength of DL-PLA is 1.35 MPa and 20% β-TCP/DL-PLA is 1.75 MPa,there is a significant difference between them (P<0 05).At the end of 12 weeks DL-PLA is 0.13 MPa,20% β-TCP/DL-PLA is 0.22 MPa.Conclusion The degradation of 20% β-TCP/DL-PLA is more suitable for bone regeneration and cell implanted and the enviroment during degradation,which is advantageous to the growth of bone regeneration.
