葛根素对缺氧-复氧时乳鼠心肌细胞分泌细胞因子的作用 被引量:18

摘要 目的 观察葛根素 (Pue)对缺氧 -复氧诱导的大鼠乳鼠心肌细胞分泌的肿瘤坏死因子 α(TNF α)及白细胞介素 6(IL 6 )的作用。方法 采用生物学方法 ,测定对照组、模型组、及 1g·L-1、0 1g·L-1、0 0 1g·L-1Pue给药组在各个时间点时细胞培养上清液中TNF α、IL 6活性的变化。结果 受到缺氧、复氧刺激后TNF α、IL 6活性较对照组增加 (P <0 0 1)。各剂量在缺氧时均表现出降低TNF α活性的作用 ,1g·L-1Pue的作用最强 (P <0 0 1) ,而其它两个剂量在复氧时还能增加TNF α活性。Pue能降低IL 6活性的增加。结论 缺氧、复氧刺激能增加心肌细胞分泌TNF α、IL 6 ;Pue能抑制缺氧时TNF α的分泌 ,但在复氧后 ,除 1g·L-1外 ,其它剂量还表现出升高TNF α的作用 ;Pue可抑制IL 6的过度分泌 ,并呈剂量依赖趋势。 AIM To investigate the effect of puerarin (Pue) on tumor necrosis factor (TNF) α, interleukin (IL) 6 secreted by neonatal rat cardiomyocytes during hypoxia/reoxygenation injury. METHOD The activities of TNF α and IL 6 in the supernatants of cultured myocytes, which were sampled from different groups (control, model, and therapeutic groups with 1 g·L -1 Pue, 0 1 g·L -1 Pue, 0 01 g·L -1 Pue) at different time, were assayed by bioassay method. RESULTS TNF α and IL 6 activity increased compared with that of control ( P< 0 01). Each dose decreased TNF α activity during hypoxia (1 g·L -1 Pue was the most potent, P< 0 01), while 0 1 g·L -1 , 0 01 g·L -1 Pue increased it during reoxygenation. Pue lowered the increase of IL 6 activity. CONCLUSION The activities of TNF α and IL 6 secreted by myocytes are enhanced by the stimulation of hypoxia and reoxygenation; Pue inhibits TNF α during hypoxia, while lower doses increase it during reoxygenation; Pue can inhibit the oversecretion of IL 6 in a dose dependent manner.
