摘要
为探讨牛磺酸对溶栓疗效的影响，测定了未溶栓和用尿激酶（ＵＫ）或ＵＫ联合牛磺酸溶栓治疗的急性心肌梗塞（ＡＭＩ）患者血浆内皮素（ＥＴ）含量的变化，并在大鼠腹主动脉血栓模型上观察用ＵＫ或ＵＫ联合牛磺酸溶栓后血浆ＥＴ、丙二醛含量以及血管内血栓变化。结果显示：（１）ＡＭＩ发病早期（６小时）血浆ＥＴ含量明显高于正常对照值（４７．３±６．３ｎｇ／Ｌ比２０．４±９．７ｎｇ／Ｌ，Ｐ＜０．００１），２天后恢复正常，用ＵＫ溶栓后２～６小时（发病６～１０小时）血浆ＥＴ比未溶栓组增高更显著（８小时７０．８±６．６ｎｇ／Ｌ比５６．９±８．６ｎｇ／Ｌ，Ｐ＜０．０１），用ＵＫ联合牛磺酸溶栓后血浆ＥＴ含量在发病８小时至１周显著低于ＵＫ组（８小时３３．３±８．２ｎｇ／Ｌ比７０．８±６．６ｎｇ／Ｌ，Ｐ＜０．０１）。（２）动物试验与临床结果相似，并发现ＵＫ联合牛磺酸治疗比单独用ＵＫ明显抑制了血管再通后丙二醛含量（２０．８５±３．０５μｍｏｌ／Ｌ比２５．１８±３．５３μｍｏｌ／Ｌ，Ｐ＜０．０５），且血栓占血管段面积比值也显著降低（０．４６５０±０．０９２８比０．６１７６±０．１１７９，Ｐ＜０．０５）。提示：牛磺酸可明显降低血浆ＥＴ含量，且能增强溶栓?
Inordertoinvestigatetheinflucencesoftaurineonthrombolysisserumedothelin(ET)concentrationwasdeterminedinpatientswithacutemyocardialinfarcation(AMI)withouturokinase(UK)treatment(group1)andaftertreatmentwithUK(group2)orUKcombinedwithtaurine(group3).InaratmodelwithabdominalaortathrombosedbyFeCl3,thechangesofserumET,malodialde-hyde(MDA)andintravascularthrombosiswereobservedinthreegroupssameasinpatients.There-sultswereasfolows:(1)SerumETlevelsofgroup1patientsatearlyphaseofonset(6hours)weresignificantlyhigherthanthoseofthecontrols(47.3±6.3ng/Lvs20.4±9.7ng/L,P<0.001).AftertwodaysserumETdecreasedtonormallevel.SerumETlevelsweresignificantlyhigherfrom6to10hoursaftertheonsetofAMIingroup2thaningroup1(70.8±6.6ng/Lvs56.9±8.6ng/L,P<0.01,at8hours).SerumETlevelsweresignificantlylowerfrom8hourstoawekafteronsetofAMIingroup3thaningroup2(33.3±8.2ng/Lvs70.8±6.6ng/L,P<0.01,at8hours).(2)Intheratmodelwiththrombosisofabdominalaorta,thechangesofserumETweresimilartothoseofAMIpa-tients.InadditionserumMDAlevelsweresignificantlydecreased(20.85±3.05μmol/Lvs25.18±3.53μmol/LaftercombinedtreatmentwithUKandtaurine,P<0.05).Theratioofcrossareaofvascu-larlumenandthrombuswasloweraftertreatmentwithcombinationofUKandtaurinethantreatmentwithUKalone(0.4650±0.0928vs0.6176±0.1179,P<0.05).theresultssuggestedthattaurinecandecresesignificantlyserumETlevels,potentiateUK-inducedvascularrecanlizationandreduceischemiareperfusioninjury.Taurinemightbeusefulclinicalyasanadjunctofthrombolytictherapy.
