摘要
目的 :探讨创伤后不同时间点脾细胞核因子 kappa B(NFκB)的 DNA结合蛋白和 NFκB p6 5蛋白亚型表达的动态变化。方法 :采用小鼠双后肢闭合性砸伤 +骨折模型 ,于创伤后 1、4、7日处死动物 ,分离、纯化脾细胞 ,提取脾细胞核蛋白。用电泳迁移率改变试验 (EMSA)检测 NFκB的 DNA结合活性 ;用免疫蛋白印迹法 (Western blot)检测 NFκB p6 5蛋白亚型表达。结果 :脾细胞 NFκB的 DNA结合活性在创伤后 1、4、7日表达明显增高。 NFκB p6 5蛋白亚型在创伤后 1、4、7日表达明显增加。结论 :创伤可明显增强脾细胞NFκB的 DNA结合活性和 p6 5蛋白亚型的表达。在创伤后淋巴细胞活化及全身炎症反应综合征 (SIRS)发生。
Objective:To observe the dynamic changes in the activation of nuclear factorkappa B( NFκB ) and the expression of p65 protein subunits of NFκB in mice spleen lymphocytes after trauma.Methods:Closed impact injury with fracture of both hind limbs of mice was adopted as trauma model.Animals were sacrificed on 1,4, and 7 days after the injury.Spleen lymphocytes were isolated and nuclear protein was extracted.DNA binding activity of NFκB was measured using electrophoretic mobility shift assay(EMSA).The expression of p65 protein subunit of NFκB was examined with Western blot.Results:DNA binding activity of NFκB in spleen lymphocytes of traumatized mice gradually increased and was significantly on day 1,4 and 7 after the injury,Western blot showed significantly increased expression of p65 protein subunits of NFκB on day 1,4 and 7 after the injury.Conclusions:These data suggest that the expression of the DNA binding activity and p65 protein subunit of NFκB significantly increased in spleen lymphocytes of traumatized mice.These changes may play an important role in mediating lymphocytes activation caused by trauma and developing of systemic inflammatory response syndrome (SIRS).
