摘要
目的 :观察白细胞介素 10 (IL 10 )对IL 1β诱导的人系膜细胞 (HMC)前列腺素E2 (PGE2 )释放和胞浆型磷脂酶A2 (cPLA2 )及环氧化酶 2 (COX 2 )基因和蛋白表达的影响。 方法 :利用体外培养的人系膜细胞 (HMC) ,设立对照组、IL 10 (2 5ng/L)处理组、IL 1β(10ng/L)处理组及IL 10 +IL 1β处理组 ;采用放射免疫测定法检测各组HMC培养上清中PGE2 ,应用RT PCR和WesternBlot检测各组cPLA2 、COX 2mRNA和蛋白水平。 结果 :正常情况下 ,HMC低水平表达cPLA2 、COX 2mRNA和蛋白并释放少量的PGE2 ;IL 1β能显著上调HMCPGE2 释放及cPLA2 、COX 2mRNA和蛋白的表达 (P均 <0 0 1) ;IL 10对基础状态下的HMCPGE2 释放及cPLA2 、COX 2mRNA和蛋白表达无明显影响 (P均 >0 0 5 ) ,但可显著下调IL 1β诱导的PGE2 释放及cPLA2 、COX 2mRNA和蛋白表达 (P均 <0 0 1)。 结论 :IL 10抑制IL 1β诱导的HMCPGE2 释放及cPLA2 和COX 2表达 ,提示IL
Objective:To investigate the effects of interleukin 10(IL 10) on IL 1β induced prostaglandin E 2 (PGE 2) release,the cytoplasmic phospholipase A 2 (cPLA 2) and cyclooxygenase 2(COX 2)expression of mRNA and protein in human mesangial cells (HMCs) and to examine IL 10 having possible effect on the biological function of IL 1β. Methodology:HMCs were cultured in vitro and divided into four groups.HMCs were treated with vehicle(group one),with IL 10 (25 ng/L)(group two),with IL 1β(10 ng/L)(group three),and with IL 10 plus IL 1β(group four)for 24 hours.The PGE 2 concentration in supernatants of HMCs was measured by radioimmunoassay.The cPLA 2 and COX 2 mRNA expressions were detected by RT PCR and the protein production was determined by Western Blot respectively. Results:HMCs expressed on low level of cPLA 2,COX 2 mRNA and protein,and released a small quantity of PGE 2 in control.The levels of cPLA 2,COX 2 mRNA and protein or PGE 2 were increased after being treated by IL 1β in HMCs ( P <0.01).IL 10 could downregulate the expression of cPLA 2 and COX 2 in HMCs induced by IL 1β,however,IL 10 had no such effects alone. Conclusion:IL 10 may reverse the release of IL 1β induced PGE 2 and expression of cPLA 2 and COX 2.It indicates IL 10 having anti inflammatory effects at several aspects by way of inhibiting the production of pro inflammatory cytokines and suppressing the biological function.
