摘要
以烟草花叶病毒 (TMV)及其抗体为材料 ,以硝酸纤维素膜 (NCM)为载体 ,根据斑点免疫吸附法 (DIBA)的原理 ,在国内首次研制出斑点免疫诊断试剂盒。研究表明 :以健康汁液预先吸附抗血清以及用 1%H2 O2 处理点样后的NCM ,能有效地排除斑点免疫吸附中非特异性干扰 ;用氯仿处理研磨后的样品和适当提高样品的稀释度能有效去除样品中叶绿素和其它色素的干扰 ,提高灵敏度。制备出的试剂盒具有体积小、重量轻、便于携带等优点 。
By using HRP goat anti rabbit IgG,nitrocellulose membrane(NCM)as solid phase and 3,3 Diamino benzidine as substrate and stain,dot immunobinding assay(DIBA)kits was developed The results indicated that the nonspecific reactions could be avoided by preparations as pre absorption of TMV antiserum with healthy tobacco leaf sap and treatment of NCM with 1% H 2O 2 for 15~20 min Increasing dilution of sample sap with PBST and treating sample sap with chloroform could remove chlorophyll and other pigments.These methods effectively reduced the background color and increased sensitivity.The simple,rapid and sensitive kits would be proved to be useful and practical for the detection and diagnosis of TMV.
