摘要
目的为证实纯化的牛泪腺活性蛋白抗原(分子量43kD或67kD)的免疫效应及其在实验性自身免疫性泪腺炎(experimentalautoimmunedacryoadenitis,EAD)制作中的作用。方法用纯化的牛泪腺抗原及非特异性抗原植物血凝素(phytohemagglutin,PHA)作为诱导,H3-TDR掺入法检测Wistar大鼠EAD淋巴细胞转化试验。结果EAD组淋巴细胞对PHA的刺激转化反应比正常对照组偏低(P>0.05),相反对牛泪腺抗原的刺激产生高水平转化。结论牛泪腺抗原具有免疫活性,可使模型鼠泪腺致敏,动物泪腺之间存在交叉反应。
ObjectiveThe study was designed to verify the immunologic effect of purified (MW 43 kD or 67 kD) bovine lacrimal gland antigen and its role in the establishment of experimental autoimmune dacryoadenitis (EAD).MethodsDEAE52 ion exchange chromatography was used to purify bovine lacrimal gland antigen,which yielded an activated protein fraction (MW 43 kD or 67 kD).Wistar rats were immunized with this purified activated protein fraction in complete Freund’s adjuvant to establish the EAD model.We performed the lymphocyte blastogenesis test by using the mixed H3TdR method and nonspecific antigen phytohemagglutin (PHA) or purified activated protein fraction as a stimulant.ResultsWistar rat EAD models were produced by immunization with the purified bovine lacrimal gland antigen.Blastogenesis tests show that the stimulation index(SI) induced by PHA in the rat model was slightly lower than that of normal controls (P>0.05).In contrast,the SI was highly induced by the purified bovine lacrimal gland antigen.ConclusionBovine lacrimal antigen has an immunologic effect in the model rat,due to a crossreaction between the antigen and the animal’s lacrimal gland.
