摘要
目的 探讨小儿急性白血病骨髓细胞Bcl- 2、P53 蛋白的表达状态。方法 以SP免疫组化法对 41例患者及 11例对照骨髓细胞Bcl- 2、P53 蛋白进行检测 ,高倍镜下计数 5 0 0个细胞 ,P53 阳性细胞≥5 %为阳性病例 ,Bcl- 2≥ 10 %为阳性病例。结果 Bcl- 2蛋白表达率在小儿ALL、ANLL及ALL高危型中均显著高于对照组 (P <0 .0 5 ,P <0 .0 0 1,P <0 .0 1) ,ALL标危型与对照组相比无差异 (P >0 .0 5 ) ,但ALL高危型高于标危型 ,ANLL高于ALL(P <0 .0 5 ,P <0 .0 5 )。P53 蛋白表达率在ALL、ANLL、ALL高危型及标危型均显著高于对照组 (P <0 .0 5 ,P <0 .0 1,P <0 .0 5 ,P <0 .0 5 ) ,ALL高危型与标危型及ALL与ANLL之间无差异 (P >0 .0 5 )。结论 P53 突变及Bcl- 2表达增强参与了小儿急性白血病骨髓细胞的恶性转化 ,但其作用机制可能是相互独立的。
Objective To explore the Bcl-2 and P 53 protein expression of bone marrow cells in children with acute leukemia (AL). Method The Bcl-2 and P 53 protein of bone marrow cells in 41 cases of acute leukemia and 11 controls were detected by SP immunohistochemical assay. By counting 500 cells in high-power microscopic fields, P 53 postive cells≥5% was postive cases and Bcl-2≥10% was postive cases. Result The expressive rate of Bcl-2 protein in children with ALL, ANLL and high risk cases of ALL was significantly higher than that of controls (P<0.05, P<0.001, P<0.01). No difference was found between standard risk cases of ALL and controls (P>0.05), but high risk cases of ALL was higher than cases of standard risk and ANLL was higher than ALL (P<0.05). The expressive rate of P 53 protein in ALL,ANLL, high risk of ALL and standard risk cases was significantly higher than that of controls (P<0.05, P<0.01, P<0.05, P<0.05). There were no significant difference between the high risk and standard risk and the ALL and ANLL patieats (P>0.05). Conclusion The Bcl-2 higher expression and P 53 mutation participates in malignant transformation of bone marrow cells in AL children, but their mechamisms of action may be independent mutually.
