摘要
目的 用恶性疟原虫FCC1/HN株CSP基因表达产物免疫小鼠 ,观察其在宿主体内各组织的抗原分布情况。方法 提取目的基因PfCSP在HeLa细胞中的表达产物 ,免疫注射BALB/c小鼠。取免疫后 4周及 7周小鼠心脏、肝脏、脑、脾及肌肉 ,通过免疫酶组织化学法及间接免疫荧光法检测组织内的抗原。结果 免疫 7周后的BALB/c小鼠肝脏组织枯否氏细胞内及肝细胞膜上可见棕黄色特异性抗原 -抗体反应 ,而免疫鼠心、脑、肌肉未出现明显的阳性反应。结论 恶性疟原虫FCC1/HN株CSP基因表达蛋白能被肝实质细胞特异性识别 。
Aim To observe the antigen distributing in the tissues of mice immunized with the expressed product of CSP gene from Plasmodium falciparum.Methods The expressed protein PfCSP in human HeLa cells was isolated and used for immunization of BALB/c mice.The presence of CSP expressed products was detected in the muscles,livers,spleens,hearts and brains of mice immunized with the condidate vaccine 4 weeks and 7 weeks by indirect immunohistochemistry assay and indirect immunofluorescent assay.Results There were the positive reactions in the cytoplasm of hepatic Kupffer cells and on the surface of hepatocytes of mice immunized after 7 weeks.No expressed products were detected in other tissues.Conclusion The expressed protein of PfCSP can specifically be distinguished by hepatic cells.There were PfCSP antigen in the livers after immunization.
