摘要
目的 对获得的日本血吸虫 (Schistosomajaponicum ,Sj)精氨酸酶基因cDNA序列进行二级结构预测。方法 采用生物信息学等技术对获得的Sj精氨酸酶编码基因序列进行开放读框 (ORF)的寻找 ,编码氨基酸的推导 ,蛋白质同源性比较以及二级结构的预测。结果 获得的cDNA序列为 10 6 1bp ,含有一个 840bp的阅读框 ,编码 2 79个氨基酸 ,属精氨酸酶家族 ,与国际蛋白质库中的酵母、蛙、人和大鼠精氨酸酶序列的同源性分别为 44 % ,5 0 % ,5 1% ,5 3%和 5 4%。Sj精氨酸酶氨基酸序列的 β转角、亲水性、平均可塑性和无规则卷曲的分布主要出现在N -末端AA残基 30~ 5 0 ,70~ 90和 180~ 2 0 0等 3个区域 ,是潜在的抗原表位。结论 推测Sj精氨酸氨基酸酶具有较好的抗原性。
Aim To predict the secondary structure of the cDNA encoding Schistosoma japonicum(Sj) arginase.Methods By using bioinformatics,the cDNA sequence encoding Sj arginase was analyzed,including searching the ORF,translating the nucleotide to protein sequence,similarity searches and secondary structure predication.Results The cDNA fragment with 1061 bp included a FRO with 840nt,and translated into 279 amino acid residues.The deduced protein was similar to the arginase family,the homology at amino acid sequence level was 44%,50%,51%,53% and 54% with Schizosaccharomyces pombe(fission yeast) arginase,Xenopus laevis(African clawed frog) arginase,Homo sapiens(human) arginase ii precursor and Rattus norvegicus(rat) arginase ii precursor respectively.The amino acid in beta-trun,hydrophobicity,flexibity and coil were possibly localized in tis N-terminal No.30~50,70~90 and 180~200,these site were probaly potential antigen epitopes.Conclusion This poly peptide of Sj arginase is considered to have stronger antigenicity.
