日本血吸虫中国大陆株32ku抗原cDNA的克隆及其核苷酸序列分析 被引量:2

摘要 目的 研究日本血吸虫重组疫苗 ,表达和制备血吸 32ku抗原重组疫苗。方法 根据Merckelbach等报道的日本血吸虫中成虫 32ku抗原完整编码序列设计引物 ,以日本血吸虫中国大陆株成虫mRNA为模板 ,采用逆转录 -聚合酶链反应技术 ,扩增其 32ku抗原完整编码区cDNA。将cDNA重组于pGEM Teasy载体上 ,转化大肠杆菌。结果 重组质粒经测序进行鉴定 ,结果与Merckelbach等报道的 32ku抗原编码区cDNA进行比较 ,二者序列一致。结论  32ku抗原在不同的地区异株间的 32ku蛋白进化上存在高度保守性 ,因此 32ku抗原可以作为重组疫苗的候选抗原。 Objective To study and construct the recombinant vaccine of Chinese mainland strain Sj32ku antigen.Method Based on the Sj32ku gene sequence published by Merckelbach, primers were designed to amplify the Sj32ku gene from the Chinese mainland strain Sj32ku using RT-PCR method. The gene was inserted into plasmid pGEM-T easy and the recombinants were transformed toE.coli DH5α. A recombinant colony has been obtained and identified by sequencing. Results The DNA sequence of the Sj32ku gene cloned from the Chinese mainland strain Sj32ku was identical to that in the Merckelbach report.Conclusion The Sj32ku gene is highly conserved among different strains and is suitable for the development of recombinant vaccine. This study should facilitate the development of the recombinant vaccine for the Chinese mainland strain.
