摘要
对一种新近分离的蜡状芽孢杆菌 (BacillusCereus)菌株S1发酵产新型抗真菌多肽APS的发酵培养基组成 (碳、氮源 )和工艺条件 (发酵温度、初始pH、通气方式和通气量 )进行了摸索 ,通过单因素实验和正交优化实验 ,确定了S1发酵培养基的组成 :麸皮 5%、玉米粉 5%、尿素 0 .2 % ,或NH4NO31 .5%、酵母浸膏 1 .5%、葡萄糖 6%、NaCl0 .1 % ;最适发酵培养温度为 2 8℃ ;最适发酵培养初始pH为 7.4或 6.8。在优化条件下 ,效价最高为 8~ 1 0mg/ml。S1生长的最适温度和初始pH为 30℃和 7.0。通气对S1发酵具有显著的影响。
The fermentation conditions of Bacillus cereus strain S1 producing a novel cyclic polypeptide (APS) were conducted by designs of single factor and orthogonal experiments.Factors such as temperature, initial pH, nitrogen sources, carbon sources etc. were studied. Two optimized fermentation media were as follows :wheat brown 5%, maize powder 5%, urea 0.2% and glucose 6%, NH 4NO 3 1.5%, Yeast extract 1.5%, NaCl 0.1%. The optimum temperature was 28℃ and the optimum initial pH was 6.8 or 7.4 . The transferred amount of oxygen had significant effects on the production of APS .
