Effects of 1,25-Dihydroxyvitamin D_3 on Bone Metabolism of Rats Exposed to Simulated Weightlessness (Skeletal Unloading)

摘要 本实验观察了1,25(OH)_2D_3对模拟失重大鼠骨质代谢的影响。结果表明,补充1,25(OH)_2D_3显著提高模拟失重大鼠肠钙吸收,增加胫骨骨形成率,和血钙、血磷浓度。对肠ALP活性和骨钙、骨磷含量作用不明显。同时观察到模拟失重大鼠肾脏MDA含量显著增加,肝脏MDA含量增加不明显。结果提示,在失重或模拟失重条件下,用1,25(OH)_2D_3可改善骨质代谢。 In order to examine the effects of 1,25-dihydroxyvitamin D_3 on the process of osteopenia following skeletal unloading, the rear limbs of rats were unloaded for 21 days. In the last 7 days, the rats were given 1, 25-dihydroxyvitamin D_3(0.03μg) orally. Control animals were given 1,2-propanediol. The results showed that skeletal unloading reduced bone formation and serum concentration of P, reduced Ca and P content in bone as well as intestinal absorption of Ca. 1,25-dihydroxyvitamin D_3 increased intestinal absorption of Ca, increased bone formation rate of tibia and serum concentration of Ca and P, significantly. Ca and P contents in femur of experimental rats did not differ from that in femur of control ones. There was no great difference in ALP activity of small intestine between control and experimental rats. It was also found in this study that the malon-dialdehyde (MDA) content in kidney of rats exposed to skeletal unloading was significantly increased but that in liver was not, and the administration of 1,25-dihydroxyvitamin D_3 did not affect MDA content in both kidney and liver of rats expesed to skeletal unloading. These results indicate that 1,25-dihydroxyvitamin D_3 can be used as one of the measures which improve bone metabolism in skeletal unloading.
