摘要
目的探讨瘦素通过对椎体骨质代谢的影响,从而进一步影响椎间盘退变的可能性。方法获取12个6月龄雄性ob/ob小鼠(瘦素缺乏小鼠)和野生型小鼠(C57BL小鼠)的腰5椎体(各6个),通过Micro-CT检测比较两者骨密度、皮质骨厚度、小梁骨体积、小梁骨厚度、小梁骨数量、连接性密度、结构模型指数等显微结构参数的差异。结果与野生型小鼠相比较,6月龄ob/ob小鼠的椎体骨密度及小梁骨厚度增大,椎体连接性密度(Conn.D)明显增加;椎体骨小梁间隙明显减少;椎体结构模型指数(SMI)明显减小,两者比较差异均有显著统计学意义(P<0.01);其小梁骨体积((BV/TV)、小梁骨数量增加、皮质骨厚度增大,两者比较差异均有统计学意义(P<0.05)。结论瘦素可能通过对小鼠椎体骨质代谢影响椎间盘的营养供应从而进一步影响椎间盘的退变。
Objective To explore the effect of leptin on bone metabolism of the vertebral body, and to further discuss the possibility affecting the intervertebral disc degeneration.Methods The bodies of the 5th lumbar vertebral were surgically obtained from 6 wild type mice ( C57BLmice) and 6 ob/ob mice ( leptin-deficiency mice) .The variation of the micro-structural parameters, including bone mineral density ( BMD ) , cortical thickness, trabecular bone volume, trabecular thickness, trabecular number, connectivity density ( Conn.D) , and the structure model index were evaluated using Micro-CT.Results Compared with those in wild type mice, significant increase of BMD of the vertebral bodies, trabecular thickness, and Conn.D was observed in 6-month ob/ob mice, while the significant decrease of the trabecular separation and the vertebral structure model index ( SMI ) were also observed in ob/ob mice (P&lt;0.01).Meanwhile, trabecular bone volume fraction (BV/TV), trabecular number, and the cortical thickness in ob/ob mice also increased compared with those in wild type mice ( P&lt;0.05 ) .Conclusion Leptin may affect the nutrition supply of the intervertebral disc through its effect on bone metabolism of the vertebra body, thus affecting the intervertebral disc degeneration.
