摘要
在 pH2.53的介质中 ,曙红Y与牛血清蛋白 (BSA)作用形成复合物 ,最大吸收峰从513nm红移到529nm ,曙红Y荧光光谱发生显著变化 ,最大发射光波长540nm(最大激发光波长308nm)荧光猝灭 ,其猝灭程度与BSA含量成正比 ,线性范围0~2.5mg/L,相关系数0.999 ,检出限2.8μg/L(3σ)。作者研究了反应条件、干扰物质及进行了样品测试 ,结果令人满意。
In a medium at pH 2.53 Eosin Y forms an adduct with bovine serum albumin(BSA), and causes a red _ shift of its maximum adsorption peak from 513 nm to 529 nm. In the meantime, there is no deformation or shift of its excitation emission peaks. The maximum fluorescence emission of Eosin Y at 540 nm(with excitation wavelength 308 nm) can be quenched upon the addition of BSA. The intensity of fluorescence quenching is directly proportional to BSA content. This quenching reaction can be used for the rapid and sensitive determination of BSA. The linear range is 0~2.5 mg/L, while the correlation coefficient is 0.999. The detection limit is 2.8 μg/L(3 σ). The tolerance towards interfering substances is satisfactory.
