摘要
将分化出的冬小麦花粉植株的苗龄分为绿芽,1cm绿苗、3cm绿苗,经3—8℃的冷库内生长66天,这些不同苗龄的绿苗均生长3cm左右,越夏后生根培养一个月,其生根率分别为96.4%、72.9%、70.6%,以绿芽越夏后生根率最高。移栽成活后处于分蘖盛期的单倍体花粉植株,用0.04%的秋水仙碱与1.5%的二甲基亚砜混合液在18℃条件下浸泡分蘖节6—8h,加倍率可达82.9-87.7%。
Age of winter wheat plantlets regenerated from anthers was called green bud, 1cm height plantlets and 3cm height plantlets. Growing for 66 days at 3—8℃, these green plantlets of different age increased about 3 cm at height. After they passed summer, they were cultured 1 month for root production. The root productive freguency was 96.4%, 72.9% and 70.6% respectivly, among which the highest was the plantlets at green bud age. At peak tillering stage of survival plants, chromosome double frequency reached from 82.9% to 87.7% by steeping tiller nodles for 6—8hr. in a mixed solution of 0.04% calchicine and 1.5% dimethyl sulfaxide.
