适宜多基因型青杨杂种叶片再生体系建立 被引量:2

摘要 以青杨杂种全同胞家系内30个基因型的叶片为外植体,研究培养基、基因型及基因型×培养基互作效应对青杨杂种叶片分化的影响,筛选适合多基因型青杨叶片分化的广谱型培养基,为对青杨从群体水平进行遗传转化和多倍体诱导奠定技术基础。研究结果表明:(1)基因型、培养基、基因型×培养基互作效应对青杨杂种叶片分化的影响极显著。(2)筛选出适合多基因型叶片分化的广谱型培养基为MS+6-BA0.4 mg·L-1+NAA 0.05 mg·L-1,可满足56.7%的青杨杂种基因型叶片分化,分化率可达70%以上。研究认为:在对群体再生生产中,应充分利用基因型×培养基互作效应,通过广谱型培养基实现大多数基因型青杨杂种叶片分化体系建立,并通过特殊培养基解决特殊基因型的青杨杂种叶片分化体系建立问题。 To study the effects of medium(E) ,genotype(G) and G x E interactions on the leaf regeneration of Populus cathayana,and to selected the suitable medium, leaves of thirty genotypes from the same full-sib family was used as explants. Related studies will provide a platform for further genetic transformation and polyploidy induction of Populus cathayana at population level. The result showed that Genotype (G), medium (E) and G × E interactions had extremely effect on shoot regeneration of hybrids of Populus. The widely-used medium of shoots induction from leaves was selected, it is the MS medium containing 0.4 mg·L^-16 -BA and 0. 05 mg·L^-1 NAA. It can induct 56.7% genotypes of Populus cathayana to generate shoots, which frequency reaches 70% o The study suggests that, tot plant regeneration in population level, G x E interactions should be take a full consideration. The widely used medium can be used for the establishment of leaf However, the special media should be regeneration system for the majority hybrids of Populus cathayana genotype selected for a certain genotype.
