内皮素-1与肝脏缺血再灌注损伤的实验研究 被引量:4

Experimental studies on the effect of endothelin 1 on hepatic ischemia reperfusion injury in rats

摘要 目的 探索内皮素 1(ET 1)在肝脏缺血再灌注损伤中的作用。方法 选用雄性Wistar大鼠 80只 ,分为正常对照组、缺血再灌注组、生理盐水组和ET 1抗体组 ,观察肝脏缺血 6 0min再灌注 3h后血浆ET 1、丙氨酸转氨酶 (ALT)、透明质酸 (HA)、以及肝组织中ET 1和丙二醛 (MDA)含量的变化 ,并观察肝组织病理学变化 ,同时 ,在缺血再灌注组选择第 1、3、6、12和 2 4h时相点观察ET 1的变化规律。结果 肝脏缺血再灌注后 ,血浆和肝组织中ET 1,血浆HA、ALT和肝组织中MDA显著升高 ,而ET 1抗体组血浆ET 1、HA、ALT与缺血再灌注组相比显著降低 (P <0 .0 1,P <0 .0 5 ) ,同时 ,肝组织的瘀血程度和损伤程度显著改善。结论 ET 1参与了肝脏缺血再灌注损伤 。 Objective To investigate the effect of endothelin 1 (ET 1) on hepatic ischemia reperfusion injury in rats.Methods Eighty male Wistar rats were randomly divided into the normal control, ischemia reperfusion, physiologic saline and endothelin 1 antibody groups and killed at 3 rd h after ischemia induced for 60?min and reperfused for 3?h. plasma and(or) liver (ET 1), glutamic pyruvic transaminase enzyme (ALT), hyaluronic acid (HA), malondialdehyde (MDA) were measured and the histopathologically hepatic alterations were observed. Meanwhile, the changes of plasma ET 1 were also observed at the time points of 1, 3, 6, 12 and 24?h in the ischemia reperfusion group. Results Plasma and liver ET 1, Plasma HA, plasma ALT and liver MDA concentrations were markedly increased after hepatic ischemia reperfusion, whereas the plasma ET 1, HA, ALT levels in ET 1 antibody group were significantly decreased compared with the ischemia reperfusion group( P < 0.01 , P < 0.05 ). At the same time, the hepatic congestion and damages were markedly improved.Conclusion ET 1 might be participated in injury of the hepatic ischemia reperfusion, which was correlated with the hepatic microcirculatory disturbances.
