HPV16阳性人宫颈鳞状细胞癌裸鼠动物模型的建立 被引量:1

摘要 目的采用在裸鼠腋部皮下移植人原代宫颈癌组织的方法,研究其在裸鼠体内的成瘤性,探讨此动物模型肿瘤的生物学特性。方法将人类乳头瘤病毒16(HPV16)阳性人宫颈鳞状细胞癌组织移植于裸鼠腋部皮下建立原代人宫颈癌裸鼠模型,成瘤后取出瘤组织传代,观察瘤体生长情况,60d待瘤长到直径1cm时处死后做病理检查;移植前和移植后用聚合酶链式反应(PCR)法检测肿瘤组织HPVDNA表达,对HPV16 L1和E6、E7基因结构进行移植前和移植后基因克隆和测序。结果接种后60d在接种部位可见结节状肿块,80d后肿瘤体积达(2.251±0.611)cm^3.瘤组织病理检查与人宫颈鳞癌一致;PCR检测结果患者HPVl6阳性,对动物模型的HPVl6L1、E6、E7基因序列进行研究,同源性在98%以上,发现了E6错义突变1个位点(T178G/D32E),E7同义突变2个位点(T843C/C94C T846/S95S),错义突变1个位点(A647G N29S),L1与标准株K02718.1比对未发现新的变位点。成功地建立了人宫颈鳞状细胞癌实体瘤动物模型。结论研究所建立裸鼠人宫颈癌组织模型成瘤率较高,操作简便,可保持人宫颈癌生物学特性,为进一步研究宫颈癌的发病机制提供了重要的科研工具。
