摘要
目的建立人卵巢癌组织移植裸鼠实体瘤肝脾转移癌模型。方法将1例人卵巢癌组织移植裸鼠,建立人卵巢癌裸鼠原代移植实体瘤模型的基础上,再将实体瘤腋下、背部皮下移植,实体瘤原位移植,实体瘤细胞移植裸鼠。观察裸鼠实体瘤生长和转移情况,称量其体重、肝、脾、子宫卵巢、瘤重及瘤的大小,并作病理、电镜检查。结果成功地建立人卵巢癌组织移植裸鼠实体瘤肝脾转移癌模型。组织学和超微结构形态均证明该实体瘤保留了原人卵巢癌、肝癌、脾癌特征。已传至第18代,传代移植成功率100%,肝脾转移100%。结论建立了人卵巢癌组织移植裸鼠实体瘤肝脾转移癌模型。通过18代传代和实验观察,肝脾转移100%。为人卵巢癌肝脾转移的研究提供了理想的动物模型。
Objective To establish a model of liver and spleen metastasis of human ovarian carcinoma transplanted subcutaneously in nude mice. Methods Tissue samples of human ovarian cancer were subcutaneously transplanted into nude mice. The solid tumors grown in nude mice were subsequently transplanted in the axillary region, back and ovary of nude mice, or tumor cells were inoculated subcutaneously in nude mice. The growth and metastasis were assessed. The body weight and the weight of liver, spleen, uterus, ovary and tumor were recorded, and the tumor size was measured. Tumor tissue samples were taken for histopathological and electron microscopic examination. Results The liver and spleen metastasis model of human ovarian carcinoma in nude mice was successfully established by subcutaneous transplantation. Histological and ultmstructural observation showed that the hepatic and splenic metastatic tumors retained the features of the original human ovarian carcinoma. The tumor model already passed 18 passages with a success rate of 100% of both transplantation and occurrence of liver and spleen metastasis. Conclusion A nude mouse liver and spleen metastasis model of human ovarian carcinoma by subcutaneous transplantation has been established successfully. The results of 18 passages of transplantation show a stable metastasis in the liver and spleen with a success rate of 100%, It provides an appropriate and useful animal model for studying the liver and spleen metastasis of human ovarian carcinoma,
