淫羊藿苷对阿尔茨海默病炎症模型大鼠神经细胞的保护作用 被引量:16

Protective effects of icariin on brain dysfunction in inflammatory rat model for alzheimer's disease

摘要 目的:用脂多糖(lipopolysaccharide,LPS)建立阿尔茨海默病(Alzheimer's disease,AD)炎症模型大鼠,观察淫羊藿苷(Icariin ICA)对该模型大鼠学习记忆及超微结构的影响。方法:经八臂迷宫训练成功的45只雄性Wistar大鼠随机分为对照组、模型组、淫羊藿苷30mg/kg组、淫羊藿苷120mg/kg组、双氯芬酸组10mg/kg组,采用侧脑室注射LPS(10μl)制作实验性AD炎症模型,并八臂迷宫测验并记录其数据。迷宫测验完成后断头取脑海马组织。应用电镜观察大鼠海马组织CA1区神经细胞超微结构变化,用SPSS统计学软件进行统计学分析。结果:⑴侧脑室注射LPS(10μl)后对大鼠一般情况观察出现了行为异常。⑵八臂迷宫测试系统分析结果:制模后与对照组相比,其它各组大鼠总记忆错误(TE)、参考记忆错误(RME)、工作记忆错误(WME)明显增多,用淫羊藿苷治疗后与模型组比较,淫羊藿苷120mg/kg组和双氯芬酸组10mg/kg组大鼠的TE、RME、WME均明显减少,淫羊藿苷30mg/kg组错误选择次数无统计学意义。超微结构观察示:对照组、海马组织线粒体比较规则,淫羊藿苷120mg/kg组、双氯芬酸组线粒体轻度受损,模型组海马神经细胞核固缩,核周隙扩张,甚至核膜结构不清,核形态极不规则。结论:侧脑室注射LPS(10μl)能成功复制AD炎症动物模型;淫羊藿苷120mg/kg组显著改善AD模型大鼠的学习记忆能力,淫羊藿苷对炎症模型大鼠海马超微结构有保护作用。 Objective:TO establish Alzheimer's disease (AD)inflammation model of rats using lipopolysaccharide(LPS). Then to explore effect of icariin the memory and ultrastructure in those rats. Methods: After successfully trained in the 8-arm maze , a total of 45 male Wistar rats were.randomly divided into 5 groups, including Control group, model group,dielofenac group( 10mg/kg·d), icariin 30mg/kg·d group, icariin 120mg/kg·d group. The AD model rats were established by injection LPS(10μl) into the lateral cerebral ventfical, then Carries on 8-arm maze to the above'animal to examine and to record its data for 15 days. Then the brain tissue of rats were removed and the ultrastructural changes of neurons were viewed under transmission electron microscope in hippocampal area CA1 in rats. Results: (1)The rats'behavior disorder showed up after being injected LPS(10pl) . (2). The results in 8-arm maze test : After being injected LPS as compared with the control group, the other rats in each group the total memory error (TE) ,reference memory errors (RME) ,working memory error (WME) significantly increased( P 〈 0.01 ) ,After treatment with the icariin compared with model group, icariin 120mg/kg·d group and the diclofenae group( 10mg/kg·d) of rats TE, RME, WME were significantly reduced. (3) The result of electron microscope showed : For the control group the chondriosomer apparenence was regular ,For the diclofenac group and ieariin 120mg/kg·d group ,chondriosomes was gently dam- aged , For the model .group and icariin 30mg/kg·d group the karyopyknosis was severe , the perinuclear cistema expanded, even the karyotheta structure was unclear, the nuclear shape was extremely irregular. Concluslons:Inflammation rat model with AD could be induced by injecting LPS into the lateral cerebral ventricle, icariin 120mg/kg·d group significantly improve learning and memory in AD model rats. icariin 30mg/kg · d group did not produce significant effect, and its mechanism may be through inhibiting the inflammation of nerve cell damage achieved.
