摘要
目的:观察补肾益智汤对老年痴呆模型大鼠学习记忆和有关神经递质的影响。方法:颈背部皮下注射D-半乳糖及侧脑室注射Aβ25-35制成老年痴呆大鼠模型,以低5g.kg-1.d-1、中10g.kg-1.d-1、高20g.kg-1.d-13个剂量的补肾益智汤灌胃治疗,通过八臂迷宫测试各组大鼠的学习记忆能力,采用比色法测定血清及脑组织中乙酰胆碱脂酶活力,荧光法测定脑组织中去甲肾上腺素(NE)、多巴胺(DA)、5-羟色胺(5-HT)含量。结果:高剂量补肾益智汤能显著减少大鼠参考记忆错误(RME)(P<0.05),对工作记忆错误(WME)有降低趋势,但无显著性差异;高剂量补肾益智汤能显著降低模型大鼠血清乙酰胆碱脂酶活力(P<0.05);中剂量补肾益智汤能显著降低模型大鼠脑组织中乙酰胆碱酯酶(AchE)活力(P<0.01),高剂量补肾益智汤能显著降低模型大鼠脑组织中乙酰胆碱酯酶(AchE)活力(P<0.001);补肾益智汤能增加脑组织中去甲肾上腺素(NE)、多巴胺(DA)含量,低剂量补肾益智汤显著增加脑组织中5-羟色胺(5-HT)含量(P<0.05);阳性药尼莫地平可显著增加模型大鼠脑组织中多巴胺(DA)、5-羟色胺(5-HT)含量(P<0.05),对去甲肾上腺素(NE)有增高趋势。结论:补肾益智汤可能通过降低乙酰胆碱脂酶活力、提高单胺类神经递质的途径实现其对老年痴呆模型大鼠的治疗作用。
Objective: Effect of BushenYizhi decoction on learning memory and Neurotransmitter in Central Nervous System in rats with Alzheimer's disease. Methods: The AD model of mice was established by the injection of Aβ25-35into the lateral cerebral ventricle and were given D-galactose, sc. The learning and memory ability of the AD model rats was detected by the 8 -arm radial maze test. The activities of acetylcholinesterase(AchE) in brain and serum were examined by the colorimetric method. The content of NE DA 5 - HT in brain were examined by fluorescent spectral photometrics method. Results :High doses of BushenYizhi decoction were able to decerease Alzheimer's disease model rats reference memory error(RME) ( P 〈 0.05 )and were able to decerease working memory error(WME). High doses of BushenYizlfi decoction were able to reduce the activities of acetylcholinesterase (AchE) in serum ( P 〈 0.05 ). Middle doses of Bushe- nYizhi decoction were able to reduce the activities of acetylcholinesterase(AchE) in brain (P 〈0.01 ). High doses of BushenYizhi decoction were able to reduce the activities of acetylcholinesterase (AchE) in brain (P 〈 0. 001 ). BushenY- izhi decoction were able to improve the content of NE and DA in brain. Low doses of BushenYizhi decoction were able to improve the content of 5 - HT ( P 〈 0.05 ). Nimodipine tablets were able to improve the content of DA and 5 - HT in brain of Alzheimer's disease model( P 〈 0.05 ). Of norepinephrlne (NE) have a higher trend. Conclusion: BushenYizhi decoction may have therapeutic effects through influence on acetylcholinesterase(AchE) and monoamine neurotransmitters in rats with AD.
