摘要
目的观察补肾益智汤对老年痴呆模型大鼠学习记忆和超氧化物歧化酶(SOD)、丙二醛(MDA)的影响。方法颈背部皮下注射D-半乳糖及侧脑室注射Aβ25-35制作老年痴呆大鼠模型,以低5[g/(kg.d)]、中10[g/(kg.d)]、高20[g/(kg.d)]3个剂量的补肾益智汤灌胃治疗,通过八臂迷宫测试各组大鼠的学习记忆能力,采用比色法测定大鼠血清中SOD活力,MDA含量。结果高剂量补肾益智汤能显著减少大鼠参考记忆错误(RME)(P<0.05),对工作记忆错误(WME)有降低趋势,但无显著性差异;高剂量补肾益智汤能显著提高大鼠血清中SOD活力(P<0.01);中、高剂量补肾益智汤能显著降低大鼠血清中MDA含量(P<0.01)。结论通过降低自由基水平可能是补肾益智汤治疗老年痴呆模型大鼠的机理之一。
Objective To investigate the effect of BushenYizhi decoction on learning memory, superoxide dismutase (SOD) and maleic dialdehyde(MDA) in Alzheimer's disease rats. Methods The AD model of mice was established by the injection of A[525-35 into the lateral cerebral ventricle and was given D--galaetose, sc . low, middle, high(5,10,20g/kg·d) doses of BushenYizhi decoction were orally administered. The learning and memory ability of the AD model rats were detected by 8 - arm radial maze test. The activity of SOD and the MDA in serum were examined by colorimetric method. Results High dose of BushenYizhi decoction could decrease reference memory error(RME) in AD rats ( P 〈 0.05 ), reduce working memory error( WME), and improve the activity of SOD (P 〈 0.01 ). Middle and high doses of BushenYizhi decoction could decrease the content of MDA( P 〈 0.01 ). Conclusion By reducing the level of free radicals may be the mechanisms in treating Alzheimer's rats with Bushenyizhi decoction.
