二母颗粒含药血清对肺腺癌A549及肝癌SMMC-7721细胞株凋亡的影响 被引量:2

Effect of serum containing Ermu particles on apoptosis of human lung adenocarcinoma A549 and hepatocellular carcinoma SMMC-7721 cells

摘要 目的:观察二母颗粒含药血清对肺腺癌A549及肝癌SMMC-7721细胞株增殖和凋亡的影响。方法:采用形态学及MTT法观察二母颗粒组(10g/kg)5%、10%、20%含药血清对A549及SMMC-7721细胞株增殖的抑制作用;通过流式细胞法观察药物对细胞凋亡的影响。结果:形态学观察表明二母颗粒含药血清使A549及SMMC-7721细胞株发生形态改变;MTT结果显示二母颗粒含药血清对A549及SMMC-7721细胞株增殖有抑制作用,且随含药血清浓度的增高以及作用时间的延长,抑制作用亦有所加强。其中20%含药血清作用于A549及SMMC-7721细胞株48h,10%含药血清作用于A549 48h,其OD值与对照组比较均有统计学差异;流式细胞技术检测结果表明20%含药血清作用于A549细胞株24h及48h,10%含药血清作用于A549 48h,与同时间段阴性对照组比较凋亡率明显升高;20%、10%含药血清作用于SMMC-7721细胞株24h及48h,对肿瘤细胞的凋亡率也有明显升高的作用。结论:二母颗粒含药血清对A549及SMMC-7721细胞株的增殖有抑制作用,其作用机制与其诱导细胞凋亡有密切的关系。 Objective: To observe the effect of serum containing Ermu particles on apoptosis of A549 and SMMC-7721 cells and explore the corresponding mechanism. Method: The antiprolifemtive effect of A.549 and SMMC-7721 cells were observed with morphological examination and MTT assay. The cell apoptosis detection was carried out by using flow cytometry. Result: Morphology of A549 and SMMC-7721 cells was changed after them were exposed to different doses of drug-centained serum (5% ,10% and 20% ) for 24 or 48 h. ;Serum Containing Ermu particles inhibited the growth of A549 and SMMC-7721 ceils significantly and the growth suppression rate presented time and dose dependent relationship. The OD value of A.549 and SMMC-7721 cells were significantly decreased than control groups when them were exposed to drug-contained serum(20% ) after 48h( P 〈 0.05 ). The OD value is also decreased when A549 ceils exposed to drug-contained serum( 10% ) for 48h ( P 〈 0.05). The Annexin V/PI double staining showed that the apoptosis rate increased induced by A549 cells exposed to drug-contained serum(20% ) after 24,48h and drug-contained serum( 10% ) after 48h( P 〈0.05). The apoptosis rate of SMMC-7721 ceils exposed to drugcontained serum( 10% ,20% ) after 24h and 48h also can be increased ( P 〈 0.05). Conclusion: Serum containing Ermu particles can inhibit the growth of A549 and SMMC-7721 cells by inducing cell apoptosis.
